Form IV: Colony Forming Units

Every team that participates in the 2018 iGEM InterLab study needs to fill in three forms. This is Form IV.

This form is part of your InterLab form submission, and will ask you questions about your colony forming units protocol. Fill this in as best as you can.

This form follows the InterLab Colony Forming Units Protocol, pages 13-14 from this URL:

If you have any questions or problems filling in this form, please email us at measurement AT igem DOT org.

Question Title

* 1. Team Name

Please enter your team name below. Please make sure it's the same as your official team name, including any hyphens or other characters.

Question Title

* 2. Starting Sample Preparation

Follow this protocol for your duplicate cultures of BBa_I20270 (Positive Control) and BBa_R0040 (Negative Control) only.

Please check off each step that you followed. If you did anything differently or extra, please note that in the "Other" box.

Question Title

* 3. Enter your dilution OD600 measurements below.

Question Title

* 4. Dilution Series Instructions

Please check off each step that you followed. If you did anything differently or extra, please note that in the "Other" box.

Question Title

* 5. Enter your colony counts for 8 x 10^4 dilution factor plates.

We expect many or all of these plates to have too many colonies to count (over 300 colonies). If you have too many colonies to count, this is considered "too numerous to count" so please enter "TNTC".

Question Title

* 6. Enter your colony counts for 8 x 10^5 dilution factor plates.

We expect many or all of these plates to have countable colonies.

Question Title

* 7. Enter your colony counts for 8 x 10^6 dilution factor plates.

We expect many or all of these plates to have countable colonies.

Question Title

* 8. Enter your colony forming unit calculations for each culture.

Please use the plate that has the best countable number (estimated 50-200 colonies) to calculate your CFU number. We would expect that to be dilution 4 based on the protocol but may be different depending on your results.

For example, you would enter the following information: 1 x 10^8 CFU/mL.

Question Title

* 9. Additional information

If you need more space to explain any adjustments to this protocol, please enter your comments in the box below.

Question Title

* 10. Feedback

Please let us know any other thoughts or comments you have about the InterLab study experience.
